Answer : Pour slightly more Agar than you would do normally - 400 ml from a Duran bottle should be used for every 10 plates. You can use a serological pipette for this if you really want to, with a minimum of 40ml per plate, but it tends to be slower. This prevents a large meniscus from forming across the surface of the plate.
If the ROTOR is not pinning uniformly across the plate increasing the pinning pressure slightly can help the Pad Head compensate for an uneven surface.
Some customers have found that using a higher volume such as 60ml has given them better and more even pinning across the plate.
