Picking / Dissection Advice

Hello pombe Colleagues,

 
After a postdoc filled with picking tetrads successfully, I’ve embarrassingly struggled to get dissections up and running in my independent group! See this link for a video of me failing to pick up this poor tetrad. The video is only 1 min long, but it could have much longer showing the same problem… It happens eventually, but much more inefficiently than in my postdoc. 
 
I’m using a nice, fresh needle from Singer. My plates have dried for 2 days and my inoculum for ~15 minutes before going to pick. Using 2% agar from Sunrise in YE5S.
 
Hardware doesn’t seem to be an issue. I used an MSM 400 in my postdoc in the LusKing Lab and a SporePlay now. Singer hardware has been amazing and their folks even provided me with a helpful guide targeted to my issue (attached, if anyone is curious!). Unfortunately, the problem persists.
 
Any suggestions you might have are welcome! I’ll take any critiques to my form! Honestly, if it’s some silly mistake in technique I’ve picked up that can be easily fixed, that would be great!

    Thank you to the many many members of the community that shared their wisdom in response to my question about picking tetrads a few days ago!

    When I have a better idea of the solution to my problem, I will share. In the meantime, I aggregated the advice I received in the attached document. Perhaps your labs may find it useful! If nothing else, the various strategies used across the community were fascinating to hear!
